Researchers have explored other ways to deliver insulin through the skin, such as gel-like lotions. But the skin is too good of a barrier, and drugs move into the body slowly. In contrast, the membrane lining the inside of the mouth is very thin, about one quarter the thickness of skin, making it a potential place for drugs to easily enter the bloodstream. So, Sabine Szunerits and colleagues wanted to see if a material they had previously developed — a polymer fiber mat that is activated by heat to release drugs — could attach to the cheek’s lining and deliver insulin.
The researchers first soaked small squares of a nanofiber mat, made from electrospun fibers of poly(acrylic acid), β-cyclodextrin and reduced graphene oxide, in a solution with insulin for three hours. Then the team applied the insulin-loaded patches onto cheek linings and corneas from pigs. Heating the material with a near-infrared laser for 10 minutes to 122F activated the material and released insulin into the two types of membranes several times faster than through skin. In addition, the researchers placed the patches in vivo inside the cheeks of three insulin-dependent pigs. The cheek linings showed no irritation or visual changes from the laser’s heat. As soon as the material was activated, the pigs’ blood sugar levels declined. The award-winning actor’s role in ‘Emma’
Before achieving super-stardom, the young actor landed the title role in Autumn de Wilde’s Emma. She had only appeared in a few films at that point and was nervous about convincing audiences of her acting abilities on the big screen.
The acclaimed director fell in love with Taylor-Joy, referring to her as his muse. He explained, “She would take all the pieces we’d given her – and just f—— nail it.”
In a Vanity Fair interview, Taylor-Joy referred to filming Emma as an “idyllic experience.” She explained, however, that it was also “one of the most difficult moments of her life.”
Surviving a devastating breakup

The relationship between Spike Lee and Rosie Perez started when Perez attended a nightclub. In an interview with Desus & Mero, she explained how at one point Lee happened to be in this nightclub as well. While there, Perez noticed that the director was having a ‘butt contest.’ This led to Perez confronting the filmmaker about it.
“He and I got in an argument, and he said, ‘Tonight is fate,’” Perez recalled. “And I was like, ‘You wish.’ He started laughing and he goes, ‘No, I’m telling you.’ He asked me if I act, and I said, ‘No.’ The next day my girlfriend called him because she wanted to hook up. He said, ‘No, I want to talk with the girl with the accent.’”
This upset Perez at the time, who snatched the phone away from her friend.
“I was like, ‘Yo, what the f***?” she remembered.
Lee would later go on to ask Perez to do an audition. Although she didn’t take it seriously at the time, she accepted the offer. Her audition would later result in her getting a gig in Lee’s Do the Right Thing.
Why Rosie Perez felt ‘violated in ‘Do the Right Thing’
In Do the Right Thing, the then up-and-coming actor shot her first nude scene. It was a situation that The Flight Attendant star felt uncomfortable with for several reasons, and it wasn’t necessarily about disrobing.
“It wasn’t really about taking off my clothes. But I also didn’t feel good about it because the atmosphere wasn’t correct,” Perez told the New York Times. “And when Spike Lee puts ice cubes on my nipples, the reason you don’t see my head is that I’m crying. I was like, ‘I don’t want to do this.’”
Although Perez later shared she didn’t mind disrobing for another movie, but that was only because the circumstances were different.
“But then I went and took my clothes off again for White Men Can’t Jump. But that was because it was totally my decision, I felt totally comfortable, the director was so cool and Woody Harrelson was like, ‘Well, whatever you want is cool with me,’” she explained. “So there I felt empowered by it. But with Do the Right Thing it was like, Now I’m the object, here’s the shot.”
At the time, Perez wished that she spoke up.
“And the reason why I cried was not so much because I felt violated as because I was angry at myself, because I wanted to say: ‘Say something! Get up!” So that’s how I felt violated. I felt like I violated myself,” she continued.
On her journey to becoming an actor, Rosie Perez was instructed to change certain things about herself. Namely the way she talked. According to Chicago Tribune, there were some who thought her accent was problematic. But the Emmy-nominee didn’t let that discourage her.

“Identifying combinations of antivirals is really important, not only because doing so may increase the drugs’ potency against the coronavirus, but combining drugs also reduces the risk of resistance,” Cherry said.
SARS-CoV-2, the virus that causes COVID-19, has infected 382 million people and led to 5 million deaths worldwide. There remains an urgent need for therapeutics to treat COVID-19, which has been amplified by the emerging threats of new variants that may evade vaccines. In response to this demand, Cherry and a team of collaborators have screened 18,000 drugs in search of antiviral activity, using live SARS-CoV-2 infection in human respiratory epithelial cells, because lung cells are the major target for the virus.
The researchers identified 122 drugs that showed antiviral activity and selectivity against the coronavirus, including 16 nucleoside analogs — the largest category of antivirals that are used clinically. Among the 16 were remdesivir, which is given by injection into a vein and has been approved by the FDA to treat COVID-19, and molnupiravir, an oral pill that was authorized for use in December.
Also among the 122 drug candidates, the researchers identified a panel of host nucleoside biosynthesis inhibitors, including the experimental drug brequinar. Nucleoside biosynthesis inhibitors work by blocking the body’s own enzymes from making nucleosides, which prevents the virus from being able to “steal” RNA building blocks and replicate. Brequinar is currently being tested in clinical trials as a COVID-19 treatment and as part of a potential combination therapy for some cancers.
Cherry and collaborators hypothesized that combining brequinar with a nucleoside analogue, such as remdesivir or molnupiravir, could work “synergistically” to create a more potent effect against the virus. Synergistic interactions occur when the total effect of two or more drugs is greater than the sum of the individual effects of each drug.
“We thought that using these nucleoside analogues while also reducing the levels of the host’s nucleoside building blocks might work together to super destroy the virus,” Cherry said. “It is really amazing that when you combine them, the virus is completely dead.”
The researchers tested the drugs in lung cells as well as in mice, and found that these combinations were highly effective against multiple strains of the coronavirus, including the delta variant. The team is now in the process of testing the drugs against omicron.
Additionally, the researchers found that Paxlovid — an oral antiviral that was also recently authorized by the FDA — could be combined with remdesivir or molnupiravir for an “additive” effect against SARS-CoV-2.
The next step would be for these drug combinations to be tested in clinical trials.
“As new strains of the virus emerge, the need for new treatments will remain critical,” said Frieman, co-principal investigator of the study. “We now know that there are a number of powerful drug combinations that have the potential to alter the trajectory of the virus.”

CANTON, Miss. (AP) — Morgan Freeman stars as a small-town sheriff in a movie that began filming Tuesday in Mississippi. Cole Hauser and Jaimie Alexander also are stars in the comedic noir thriller “The Minute You Wake Up Dead.”
It’s among four movies being shot this month around the state, with two more scheduled in March, according to the Mississippi Film Office.
Those two include “Broken Ties,” which will follow “The Minute You Wake Up Dead” into Canton, a city of about 11,000 about 20 miles (32 kilometers) north of Jackson, according to the film office website.
Producer Andrew Stevens told WLBT-TV that Canton was a natural fit for “The Minute You Wake Up Dead,” particularly with state subsidies.
“The small town and historical beauty perfectly fits the screenplay,” he said in an email Tuesday to The Associated Press.
He said the city’s old post office is the sheriff’s office, and its upstairs is the set for the real estate office run by Hauser’s character, Russ.
“The Diner set from ‘A Time to Kill’ is the perfect location for our movie Diner where Delaine, Jaimie Alexander’s character, works,” he wrote.
Stevens said his entire focus is on that movie, and it’s far too early to talk about “Broken Ties.”
Stevens’ family is from Mississippi — his mother, actress Stella Stevens, is from Yazoo City — and Freeman lives in Clarksdale, Mississippi.
Casting director Matthew Morgan is a native of Canton. He got his start in the industry as a production assistant and extra alongside Octavia Spencer in the movie “A Time To Kill,” which also was filmed in Canton. He is still looking for extras for “The Minute You Wake Up Dead.”
“I moved to Toronto, to Canada to be an actor,” Morgan told WLBT. “I left Mississippi to follow my dreams and ended up becoming a casting director there for probably 15 years, and then realized I could do what I love back home in Mississippi. As the industry really started thriving, I started coming back here.”
Filming for “The Minute You Wake Up Dead” winds up March 3.

De krimp is het gevolg van de hoge energieprijzen, zodat tuinders belichte teelten overslaan. Met name de beschikbaarheid van tomaten wordt zwaar getroffen. In januari werden maar liefst 50% minder losse tomaten aangeleverd bij de veilingen dan een jaar eerder.
Door het krappe aanbod stegen de prijzen die inkopers betalen. Toch kunnen daarmee de gestegen kostprijzen daarmee niet goed gemaakt worden. De kostprijzen van tuinders zijn gestegen met circa 80%, terwijl hun verkoopprijzen met 12-13% stijgen. De gestegen kosten zijn niet alleen het gevolg van gestegen energieprijzen. Ook kunstmest, arbeid en verpakkingen zijn duurder geworden. Daardoor wegen de hogere prijzen niet op tegen de hogere kosten en laten tuinders hun kas liever leegstaan.
De cijfers gelden voor in het noorden van Europa geteelde vruchtgroenten. Het leeuwendeel van de tomaten, paprika’s en komkommers die nu in onze streken in de winkels liggen komen ‘s winters uit Zuid-Europa en Marokko waar ze niet in kassen geteeld hoeven te worden.
